چکیده
By JAMES G. WmTE, WILLIAM KRIVIT AND ROBERT VERNIER T HE PRIMARY PURPOSE of the complex mechanism of coagulation is to convert soluble fibrinogen molecules into a stable meshwork of fibrin. Appreciation of the macro-molecular architecture of fibrin is essential to an understanding of how clot formation fulfills its role in hemostasis. An electron microscopic study, combining the use of immune methods, has been applied to the ultrastructural evaluation of fibrin formation. Ferritinlabelled antihuman fibrinogen antibody ( fer-A.F. antibody) incubated with plasma during clot formation specifically stains the developing fibrin strands. Periodic bands, indicative of a high degree of order, are produced by the ferritin conjugated antibody on the intrinsic cross striations of fibrin. These observations suggest the mode by which fibrinogen molecules unite to form the fibrin strand.

The Platelet-fibrin Relationship in Human Blood Clots: an Ultrastructural Study Utilizing Ferritin-conjugated Anti-human Fibrinogen Antibody.
T HE RELATIONSHIP of platelets and fibrin has interested investigators since the early descriptions of human blood clots.’4 Many studies showed an intimate platelet-fibrin association which indicated that platelets served as centers of clot formation, and as the main support of the fibrin meshwork.5’#{176} Other reports, however, described the relationship to be accidental; that is, platelets a...

Radiolabeled antibodies were perfused into fibrin clots and fibrinogen gels formed in vitro to assess the reactivity of selected epitopes. An antifibrinogen monoclonal antibody (MoAb) (antibody 1D4/xl-f), directed against an epitope in the A alpha-chain C-terminal region (A alpha 241-476), bound to 35% of the epitope in crosslinked fibrin clots and 37% of the same epitope in factor XIII-induced...
متن کاملUltrastructural changes to rabbit fibrin and platelets due to aspartame.
The coagulation process, including thrombin, fibrin, as well as platelets, plays an important role in hemostasis, contributing to the general well-being of humans. Fibrin formation and platelet activation are delicate processes that are under the control of many small physiological events. Any one of these many processes may be influenced or changed by external factors, including pharmaceutical...
متن کاملFibrinogen Cleveland II. An abnormal fibrinogen with defective release of fibrinopeptide A.
An abnormal fibrinogen (fibrinogen Cleveland II) was detected in the plasma of a 23-yr-old white man with a mild bleeding diathesis. The one-stage prothrombin time, thrombin time, and Reptilase time were all prolonged. 16 of 24 tested relatives had the defect, which appeared to be transmitted as an autosomal dominant characteristic. The thrombin time of normal plasma was slightly inhibited by t...
متن کاملRegulation by alpha 2-antiplasmin and fibrin of the activation of plasminogen with recombinant staphylokinase in plasma.
The effects of alpha 2-antiplasmin and fibrin on the activation of plasminogen by recombinant staphylokinase (STAR) were studied in an effort to elucidate further the molecular basis of the fibrin-specificity of this fibrinolytic agent. In purified systems consisting of 1.5 mumol/L intact or low-M(r) plasminogen and 3 mumol/L alpha 2-antiplasmin, at 37 degrees C and in the absence of fibrin, ST...
